Yes, it is: You can use the "Add Shape…" option and draw e.g. a box or circle where the design will be located. Then fill the shape with a Tatami pattern and make sure this object is stitched before the design (either as a separate file or by changing the thread order).
The StitchBuddy Pro license is required to save changes. I'd recommend to watch these video tutorials to make yourself familiar with adding shapes - its capabilities and limitation, and you should definitely make a test run, first.
